How to calculate platelet count manually

In this era of technology, lab technicians use CBC machine to count platelet count, but sometimes we have doubts about the CBC machine, so we make a slide and count it manually under a microscope.

So first of all we will make a slide to count the platelet count manually 

To make a slide, take a clean slide, put 5 micron of blood on the slide, slip it well and leave the slide for 5 to 10 minutes to dry.


When the slide dries completely, put Leishman Staining Solution on it and leave it for 3 minutes. After that, put buffer and if there is no buffer, then you can put distilled water on it.


Manually Platelet Count

After that, leave it like this for 10 minutes. After 10 minutes, clean it thoroughly with distilled water and then leave it to dry. When the slide is completely dry, then you can count the platelets in the microscope.


To perform a manual platelet count, you need to count the platelets in 10 fields and then add up the total number of platelets observed across these 10 fields. After that, use this method to calculate the final platelet count.

Count platelets in 10–20 fields.

Calculate the average number of platelets per field.

Multiply the average by 10000 to estimate the platelet count per µL.


For example:


If you have a total of 210 platelets counted across multiple fields, and you use a multiplier of 10,000


Platelet Count=210×10,000=2,100,000/μL


The estimated platelet count is 2,100,000/µL.
